Step 1: iPSC/ESC maintainance
day 0 ~ 1
culture “adherent” human PSC in “KnockOut DMEM” for “1” days. Medium should contain factors shown in the following table, and should be changed once in “1” days.
| concentration | unit | |
|---|---|---|
| KSR | 20 | % |
| PSA | 1 | % |
| Glutamine | 0.001 | mol/l |
| NEAA | 1 | % |
| 2-mercaptoethanol | 1.0e-4 | mol/l |
| FGF | 5.0e-6 | g/l |
Ideally, “PSC” should be obtained at the end of culture.
Step 2: Primitive streak 1
day 1 ~ 3
culture “spheroid” human PSC in “Stemline II Hematopoietic Stem Cell Expansion Medium” for “2” days. Medium should contain factors shown in the following table, and should be changed once in “2” days.
| concentration | unit | |
|---|---|---|
| streptomycin | 1 | % |
| Glutamine | 0.001 | mol/l |
| VEGF | 5.0e-5 | g/l |
| BMP4 | 5.0e-5 | g/l |
| CHIR90021 | 6.0e-6 | mol/l |
Step 3: mesoderm 1
day 3 ~ 5
culture “spheroid” human PSC in “Stemline II Hematopoietic Stem Cell Expansion Medium” for “2” days. Medium should contain factors shown in the following table, and should be changed once in “2” days.
| concentration | unit | |
|---|---|---|
| streptomycin | 1 | % |
| Glutamine | 0.001 | mol/l |
| VEGF | 5.0e-5 | g/l |
| BMP4 | 5.0e-5 | g/l |
| SCF | 2.0e-5 | g/l |
| FLT3L | 2.0e-5 | g/l |
| TPO | 2.0e-5 | g/l |
Ideally, “mesoderm” should be obtained at the end of culture.
Step 4: mesoderm 2
day 5 ~ 6
culture “spheroid” human mesoderm in “Stemline II Hematopoietic Stem Cell Expansion Medium” for “1” days. Medium should be changed once in “1” days.
Step 5: endothelial 1
day 6 ~ 8
culture “adherent” human PSC in “STEMdiff APEL2 Medium” for “2” days. For ECM, use “FN1” at “N/A” “N/A”. Medium should contain factors shown in the following table, and should be changed once in “2” days.
| concentration | unit | |
|---|---|---|
| FBS | 5 | % |
| Glutamine | 0.001 | mol/l |
| VEGF | 5.0e-5 | g/l |
| FGF | 1.0e-5 | g/l |
| DAPT | 1.0e-5 | mol/l |
Ideally, “EC” should be obtained at the end of culture.
