Step 1: iPSC/ESC maintainance

day 0 ~ 1 (duration missing in database; assumed 1 day)

culture “adherent” mouse ESC in “DMEM/F12”. For ECM, use “Fibroblast” at “N/A” “N/A”. Medium should contain factors shown in the following table.

concentrationunit
FBS15%
NEAA1%
GlutaMAX1%
2-mercaptoethanol1.0e-4mol/l
LIF1.0e+6U/l
PSA——

Ideally, “ESC” should be obtained at the end of culture.

Step 2: mesoderm 1

day 1 ~ 7

culture “spheroid” unknown species cell and mouse ESC in “unspecified medium” for “6” days. Medium should be changed once in “6” days.

Ideally, “mesoderm” should be obtained at the end of culture.

Step 3: endothelial 1

day 7 ~ 12

culture “adherent” unknown species cell and mouse ESC in “alpha-MEM” for “5” days. For ECM, use “Gelatin” at “0.1” “%”. Medium should contain factors shown in the following table, and should be changed once in “5” days.

concentrationunit
FBS10%
SCF1.0e-4g/l
IL-31.0e-4g/l
Flt3L1.0e-4g/l
DAPT2.0e-5mol/l
DMSO0.2%

Ideally, “EC” should be obtained at the end of culture. Ideally, “HE” should be obtained at the end of culture. Ideally, “LSK” should be obtained at the end of culture.
