Step 1: endothelial 1
day 0 ~ 1
culture “adherent” human PSC in “human endothelial serum-free medium” for “1” days. For ECM, use “laminin 511-E8” at “0.01” “g/m2”. Medium should contain factors shown in the following table, and should be changed once in “1” days.
| concentration | unit | |
|---|---|---|
| bovine platelet-poor plasma derived serum | 1 | % |
| FBS | 2 | % |
| FGF | 2.0e-5 | g/l |
| retinoic acid | 1.0e-5 | mol/l |
Ideally, “EC” should be obtained at the end of culture.
Step 2: endothelial 2
day 1 ~ 2 (duration missing in database; assumed 1 day)
culture “adherent” human PSC in “human endothelial serum-free medium”. For ECM, use “laminin 511-E8” at “0.01” “g/m2”. Medium should contain factors shown in the following table.
| concentration | unit | |
|---|---|---|
| bovine platelet-poor plasma derived serum | 1 | % |
| FBS | 2 | % |
Ideally, “EC” should be obtained at the end of culture.
