Step 1: iPSC/ESC maintainance
day 0 ~ 1 (duration missing in database; assumed 1 day)
culture “adherent” human PSC in “DMEM/F12”. For ECM, use “Fibroblast” at “N/A” “N/A”. Medium should contain factors shown in the following table.
| concentration | unit | |
|---|---|---|
| KSR | 20 | % |
| NEAA | — | — |
| Glutamine | 0.002 | mol/l |
| 2-mercaptoethanol | 1.0e-4 | mol/l |
| FGF | 2.0e-5 | g/l |
| PSA | 1 | % |
Ideally, “PSC” should be obtained at the end of culture.
Step 2: mesoderm 1
day 1 ~ 3
culture “adherent” human PSC and unknown species cell in “alpha-MEM” for “2” days. For ECM, use “type IV collagen” at “0.005” “g/l”. Medium should contain factors shown in the following table, and should be changed once in “2” days.
| concentration | unit | |
|---|---|---|
| FBS | 10 | % |
| FGF | 1.0e-5 | g/l |
| 2-mercaptoethanol | 5.0e-5 | mol/l |
| PSA | 1 | % |
| Activin A | 6.0e-6 | g/l |
| BMP4 | 1.0e-5 | g/l |
Ideally, “mesoderm” should be obtained at the end of culture.
Step 3: endothelial 1
day 3 ~ 8
culture “adherent” human PSC and unknown species cell in “alpha-MEM” for “5” days. For ECM, use “type IV collagen” at “0.005” “g/l”. Medium should contain factors shown in the following table, and should be changed once in “5” days.
| concentration | unit | |
|---|---|---|
| FBS | 10 | % |
| FGF | 1.0e-5 | g/l |
| 2-mercaptoethanol | 5.0e-5 | mol/l |
| PSA | 1 | % |
| VEGF | 5.0e-5 | g/l |
Ideally, “EC” should be obtained at the end of culture.
