Step 1: mesoderm 1
day 0 ~ 4
culture “adherent” mouse ESC in “alpha-MEM” for “4” days. For ECM, use “Gelatin” at “N/A” “N/A”. Medium should contain factors shown in the following table, and should be changed once in “4” days.
| concentration | unit | |
|---|---|---|
| FBS | 10 | % |
| 2-mercaptoethanol | 0.0055 | mol/l |
Ideally, “mesoderm” should express “KDR” at the end of culture.
Step 2: endothelial 1
day 4 ~ 7
culture “adherent” mouse mesoderm in “alpha-MEM” for “3” days. For ECM, use “Gelatin” at “N/A” “N/A”. Medium should contain factors shown in the following table, and should be changed once in “3” days.
| concentration | unit | |
|---|---|---|
| FBS | 10 | % |
| 2-mercaptoethanol | 0.0055 | mol/l |
| VEGF | 5.0e-5 | g/l |
| cAMP | 5.0e-4 | mol/l |
Ideally, “EC” should express “PECAM1”, “VWF” at the end of culture.
