Step 1: iPSC/ESC maintainance
day 0 ~ 1 (duration missing in database; assumed 1 day)
culture “adherent” human PSC in “unspecified medium”.
Step 2: endothelial 1
day 1 ~ 8
culture “spheroid” human PSC in “unspecified medium” for “7” days. Medium should be changed once in “7” days.
Ideally, “LEC” should express “PROX1”, “LYVE1”, “VEGFR3”, “PODOPLANIN” at the end of culture.
Step 3: endothelial 2
day 8 ~ 22
culture “adherent” human PSC in “alpha-MEM” for “14” days. Medium should contain factors shown in the following table, and should be changed once in “14” days.
| concentration | unit | |
|---|---|---|
| FBS | 5 | % |
| VEGF | 5.0e-5 | g/l |
| VEGF-C | 1.0e-4 | g/l |
| EGF | 1.0e-5 | g/l |
Ideally, “LEC” should express “PROX1”, “LYVE1”, “VEGFR3”, “PODOPLANIN” at the end of culture.
