Step 1: iPSC/ESC maintainance
day 0 ~ 1 (duration missing in database; assumed 1 day)
culture “adherent” human ESC in “mTeSR”. For ECM, use “Matrigel” at “N/A” “N/A”.
Ideally, “ESC” should be obtained at the end of culture.
Step 2: mesoderm 1
day 1 ~ 4
culture “adherent” human ESC in “DMEM/F12” for “3” days. For ECM, use “Matrigel” at “N/A” “N/A”. Medium should contain factors shown in the following table, and should be changed once in “3” days.
| concentration | unit | |
|---|---|---|
| B27 supplement without vitamin A | 1 | x |
| GlutaMAX | 1 | % |
| PSA | 1 | % |
| MTG | 4.0e-4 | mol/l |
| Ascorbic Acid | 0.05 | g/l |
| BMP4 | 2.5e-5 | g/l |
| CHIR90021 | 3.0e-6 | mol/l |
| Y27632 | 5.0e-6 | mol/l |
Ideally, “CVPC” should express “SSEA1”, “MESP1”, “MEF2C”, “GATA4”, “ISL1”, “NKX2-5” at the end of culture.
