Step 1: endothelial 1
day 0 ~ 4
culture “spheroid” human EC in “DMEM/F12” for “4” days. Medium should contain factors shown in the following table, and should be changed once in “4” days.
| concentration | unit | |
|---|---|---|
| B27 | — | — |
| SU5416 | — | — |
Ideally, “EC” should be obtained at the end of culture.
Step 2: endothelial 2
day 4 ~ 19
culture “spheroid” human EC and human Pericyte/SMC in “DMEM/F12” for “15” days. “EC” should be “upregulated” for “ID3”. For ECM, use “collagen; Matrigel” at “N/A” “N/A”. Medium should contain factors shown in the following table, and should be changed once in “15” days.
| concentration | unit | |
|---|---|---|
| B27 | — | — |
Ideally, “EC” should express “CD34” at the end of culture. Ideally, “Pericyte/SMC” should express “ACTA2” at the end of culture.
