Step 1: iPSC/ESC maintainance
day 0 ~ 1
culture “adherent” human PSC in “StemMACS iPSC-Brew XF medium” for “1” days. “PSC” should be “unspecified regulation” for “DKC1”. For ECM, use “Matrigel” at “N/A” “N/A”. Medium should contain factors shown in the following table, and should be changed once in “1” days.
| concentration | unit | |
|---|---|---|
| EDTA | 5.0e-4 | mol/l |
| Y-27623 | 1.0e-5 | mol/l |
Ideally, “PSC” should be obtained at the end of culture.
Step 2: mesoderm 1
day 1 ~ 5
culture “unspecified method” human PSC in “DMEM low glucose” for “4” days. Medium should contain factors shown in the following table, and should be changed once in “4” days.
| concentration | unit | |
|---|---|---|
| MCDB-201 medium | 40 | % |
| linoleic acid-BSA | 0.25 | x |
| ITS | 0.25 | x |
| PSA | 1 | % |
| Ascorbic Acid | 1.0e-4 | mol/l |
| Dexamethasone | 0.0025 | mol/l |
| 2-mercaptoethanol | 0.05 | mol/l |
| BMP4 | 2.0e-5 | g/l |
Ideally, “mesoderm” should be obtained at the end of culture.
